Treatment protocol |
Metformin was orally administered via drinking water at a dose of 2.5 mg/mL for 6 to 11 weeks-old, and 5 mg/mL for 12 to 20 weeks-old AS mice. Losartan was co-administered via drinking water at a dose of 125 µg/mL for 6 to 11 weeks-old, and 250 µg/mL from 12 to 20 weeks-old AS mice. The water containing these compounds was replenished twice a week.

Following fragmentation, 5.2 ug of ssDNA were hybridized for 16 hr at 45C on Clariom S Array Chips were washed and stained in the Affymetrix Fluidics Station 450.

GeneChips were scanned using the GeneChip Scanner 3000 7G.

The data were analyzed with Transcriptome Analysis Console 4.0 (TAC4.0) using Affymetrix default analysis settings and global scaling as normalization method.
