tissue: spleen
developmental stage: 8 weeks
Extracted molecule polyA RNA
Extraction protocol Total RNA from at least 50,000 isolated mouse B cells was extracted by Trizol (Life Technology) and precipitated by RNase-free glycogen(Life technology). Immediately, we isolated mRNA from total RNA with NEBNext® Poly(A) mRNA Magnetic isolation Module(E7490S).
high throughput sequencing library was constructed with NEBNext® Ultra™ II RNA Library Prep Kit for Illumina®(E7770S).

Library strategy RNA-Seq
Library source transcriptomic
Library selection cDNA
Instrument model Illumina NovaSeq 6000

Data processing Basecalls performed by Annoroad Co. Ltd. (naïve B and Day2 B) / BerryGenomics Co. Ltd. (Day3 B and day3 PCs)
RNA-Seq data were mapped by STAR aligner v2.5.2b and quantitified by Cufflinks and Cuffdiff.
Genome_build: GENCODE M12 (mm10)
Supplementary_files_format_and_content: The transcription levels (RPKM) were quantified with Cufflinks (v2.0.2) software.
