Treatment protocol |
MNT-1 melnaoma cells were serum depleted for 48 hours perior to treatment with 20uM Forskolin for 30 minutes, followed by replacment of medium with serum depleted medium. RNA was extracted at the indicated time points following stimulaiton.

MNT-1 melanoma cells were grown in 10% serum DMEM.

Extraction protocol |
RNA was isolated from MNT-1 cells using Trizol (Qiagen), according to the manufacturer’s instructions. All samples were treated with TURBO DNA-free (Ambion) to remove trace amounts of genomic DNA.

Single-stranded cDNA was generated from the amplified cRNA with the WT cDNA Synthesis Kit (Affymetrix) and then fragmented and labeled with the WT Terminal Labeling Kit (Affymetrix).

Samples were hybridized with Clarioum S Human Arrays (Affymetrix) and scanned at the HUJI Microarray Core Facility.
