Title |
Hipp IR+MSC 3 |
Sample type |
RNA |
Source name |
Hipp from mouse 3 receiving cranial radiation and MSC
Organism |
Mus musculus |
Characteristics |
tissue: hippocampus treatment: radiation and MSCs
Treatment protocol |
Frozen tissues were used for RNA isolation
Growth protocol |
Hipp and PLv from mouse brains were isolated and immediately frozen in liquid nitrogen and stored at -80 degrees C.
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA was extracted using the RNAeasy Mini kit (Qiagen) following the instructions of the manufacturer.
Label |
biotin
Label protocol |
Biotinylated cDNA were prepared according to the standard Affymetrix protocol from 2ng total RNA Labeling Protocol (Manual Target Preparation for GeneChip® Whole Transcript (WT) Expression ArraysWT Pico Reagent Kit, ©2015 Affymetrix Inc.)
Hybridization protocol |
Following fragmentation, 2.3 µg of single-stranded cDNA were hybridized for 16 hr at 45ºC on Clariom™ S Assay Mouse Array. GeneChips were washed and stained in the GeneChip® Fluidics Station 450 according to GeneChip® Expression AnalysisTechnical Manual, Affymetrix, 2004.
Scan protocol |
Microarrays were scanned using GeneChip® Scanner 3000 7G
Description |
Gene expression data from the Hipp isolated from mice receiving radiation and MSCs
Data processing |
The .CEL files were generated using Affymetrix AGCC software and further processed into expression values using Robust Multiarray Average (RMA) method by Irrizarry et al. The following statistical data analysis (ANOVA unpaired one-way) were performed using the TAC software (Transcriptome Analysis Console) developed by Affymetrix.
