Male, 11 month old mice (either wild-type or Coco/KO), were sacrificed, and then the soleus muscles excised and snap frozen in liquid nitrogen.

| Extraction protocol |
Whole soleus muscles were excised from recently euthanased mice before snap freezing in liquid nitrogen. Each soleus was cut into cryostat sections (10 microns) which were kept frozen in a dry ice-chilled tube until the addition of 1ml of TriReagent (Sigma). The TriReagent protocol was followed, except that instead of resuspending the RNA pellet into water as a final step, the pellet was resuspended into 600 microlitres of RLT buffer from the RNeasyPlus kit (Qiagen), and then the manufacturer's instructions for this kit followed from there.

Data normalization (as per Lin, Du, Huber & Kibbe (2008), Nucleic Acids Res 36, e11) and differential gene expression analysis (Bioconductor lumi and Limma software packages) was performed by Creative Biomics (http://www.creative-biomics.com/)
