Ileum were removed, flash frozen in liquid nitrogen, and RNA was harvested using Total RNA Miniprep Purification Kit (GeneMarkbio,Cat# TR01).
cDNA was generated using random N6 primer followed by A-tailing. The adaptors were ligated to the A-tailed cDNA, then were amplified by PCR. The PCR products were heated to separate into single strand DNA which was cyclized with splint oligonucleotides to generate libraries.
