Semen was collected then treated with Somatic Cell Lysis Buffer (0.1% sodium dodecyl sulfate [SDS], 0.5% Triton X-100) on ice for 30 minutes. Lack of somatic cell contamination and sperm count were established by microscopic examination. RNA was isolated from sample using Trizol. Samples were then treated with RNase-free DNase I

Biotinylated cRNA were prepared according to the standard Illumina protocol from total RNA (Whole-Genome Gene Expression with IntelliHyb Seal System Manual, 2006, Illumina).
