Growth protocol |
The human colon adenocarcinoma cell line HT29 ZsGreen/Luc+ was maintained in RPMI-1640 medium supplemented with 10 %fetal bovine serum, 1 % L-Glutamine and 1 % pen/strep antibiotic solution (growth medium) in humidified atmosphere at 37 °C in 5 % CO2. HC, CRC, HL and CRLM decellularized matrices were then injected with 2.5x105 HT-29 ZsGreen/Luc+ cells, resuspended in 10μL of Collagen I (diluted 2:3 with RPMI-1640). Complete medium was carefully added and changed every day

Total RNA was extracted from cells, HL and CRLM recellularized matrices using TissueLyser (Qiagen) combined with Trizol in 3 independent experiments

Labeled and Fragmented ss-cDNA were prepared according to the standard GeneChip WT Plus protocol starting from 100 ng of Total RNA

Hybridization protocol |
Following fragmentation and labelling,5.5 μg ss-cDNA were hybridized for 16 hr at 45C on GeneChip Human Genome Clariom S Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

GeneChips were scanned using the Scan GCS3000.
