Treatment protocol |
The cells were treated with IL-1b (10ng/ml) either with vechicle control (DMSO) or I-BET762 (500nM) for 48 h in complete media.

Growth protocol |
HPAECs were cultured in endothelial cell growth medium from Lonza. All the experiments were done in the cells with passage number 4-8.

Total RNA was isolated using Qiazol extraction method from Qiagen according to manufacturer's instructions.

Biotiny end labeled ss-cDNA were prepared according to Affymetrix WT pico protocol from 4 ng total RNA (GeneChip WT pico Reagent kit P/N 703262 rev 4).

Hybridization protocol |
Following fragmentation and end label, 2.4 ug of ss-cDNA were hybridized for 16 hr at 45C on GeneChip Human Clariom S. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

Scan protocol |
GeneChips were scanned using the GeneChip Scanner with 7G upgrade.
