Treatment protocol Semi-confluent cells were exposued to 1uM of CTD or vehicle (0.1%DMSO) dossolved in medium for 24 hours.
Growth protocol Human neuroblastoma SH-SY5Y cells (ECACC, No. 94030304) were cultured in Dulbecco’s Modified Eagle’s Medium (DMEM) with 10% fetal bovine serum (FBS), 100 U/ml penicillin and 100 µg/ml streptomycin at 37 °C in humidified air with 5% CO2.
Extracted molecule total RNA
Extraction protocol RNA was extracted from cells using a NucleoSpin® RNA isolation kit (MACHEREY-NAGEL GmbH) along with on-column DNase I treatment. RNA quality was analyzed using a Bioanalyzer 2100 (Agilent Technologies). RNA samples with RIN (RNA integrity number) values >5.0 were considered acceptable.
Label biotin
Label protocol 500 ng of total RNA was used to synthesize cRNA with a GeneChip WT PLUS Reagent Kit (Affymetrix).
 
Hybridization protocol After fragmentation, the biotinylated cRNA was hybridized to arrays at 45°C for 16 h.
Scan protocol The arrays were washed and stained in the Affymetrix Fluidics Station 450.
Data processing The data were analyzed with RMA using Affymetrix default analysis settings and global scaling as normalization method.
