Growth protocol |
For microarray transcriptome profiling of LPS tolerized human monocytes restimulated with LPS for 3, 6 and 24h. Human monocytes were isolated from buffy coats and cultured in DMEM media containing antibiotics (100IU/ml penicillin and 100ug/ml streptomycin; Invitrogen) for 2h. Non-adherant cells were removed by washing and cells were thereafter cultured in DMEM media containing antibiotics (100IU/ml penicillin and 100ug/ml streptomycin; Invitrogen) and 10% heat inactivated fetal calf serum (Pasching, Austria). These cells were used for all subsequent treatments described above.

Data processing |
Data were normalized with cross-correlation method and differential expression was based on the mean of the log2 fold changes.
