QIAGEN PAMM030 RT2 Profiler PCR Arrays were used to determine the genes altered in the insulin signaling pathway. Fast SYBR Green master max (ThermoFisher) was used during the reaction. Qiagen’s Data Analysis software was used to analyze results and construct a heatmap. All genes are normalized to 5 housekeeping genes within the array

Qiagen’s Data Analysis software was used to analyze results and construct a heatmap. Lysates were collected from 5 treated lungs (4 lungs for p16 KO room air group) and were averaged. All genes are normalized to 5 housekeeping genes within the array For the normalization it uses the average of five housekeeping genes: B2M, HPRT1 , RPL13A, GAPDH, ACTB. Values from the different treated lungs were averaged within Target gene signals normalized to housekeeping genes; 2^-deltaCt, where deltaCt = (Ct_Target − Ct_HKG)] The web-based software package automatically performs all deltadeltaCt based fold-change calculations from the uploaded raw threshold cycle data. Fold Change worksheet reports test/control (i.e.,Room air wt/ experimental CS) ratios.
