Title |
Tissue spatial point 4, biological rep2 |
Sample type |
RNA |
Source name |
Zebrafish embryos paraxial mesoderm
Organism |
Danio rerio |
Characteristics |
tissue: center PSM genotype: wild-type AB age: 12-somite stage
Treatment protocol |
Embryos were manually dechorionated at room temperature (24C°). Selected embryos were transferred to a basket in L15 media. Embryos were manually dissected and the tissue sections were placed in the Trizol solution (Invitrogen) and frozen immediately on dry ice and stored at -80C°.
Growth protocol |
Wild type embryos from AB line were grown at 23C° untill 12-somite stage.
Extracted molecule |
total RNA |
Extraction protocol |
Trizol extraction of total RNA was performed.
Label |
biotin
Label protocol |
Biotinylated cRNA were prepared by using Affymetrix GeneChip Expression 3'-Amplification Reagents for IVT Labeling kit.
Hybridization protocol |
Each chip was hybridized to 13 μg fragmented cRNA. Arrays were hybridized according to the manufacturer’s instructions.
Scan protocol |
GeneChips were scanned with a Affymetrix GeneChip Scanner 3000 7G at the Microarray Core Facility of the Stowers Institute for Medical Research
Description |
Gene expression data from the fourth spatiotemporal position along the axis
Data processing |
The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings and global scaling as normalization method. The trimmed mean target intensity of each array was arbitrarily set to 150.
