Extraction protocol |
A part of the muscle tissue was then placed in RNAlater RNA Stabilization Reagent (Qiagen, Inc., Valencia, CA, USA) and was kept overnight at 4°C and subsequently stored at 20°C. RNA was isolated using an RNeasy Mini Kit (Qiagen) and was purified according to the manufacturer’s instructions. RNA purity and concentration were evaluated with a spectrophotometer using an absorbance ratio of 260/280 nm.

The total RNA of each group was pooled (n = 10 for each group) and labeled with Cyanine 3 (Perkin Elmer, Boston, MA, USA) using 500 ng of total RNA, according to the manufacturer’s instructions, which generated one-color labeled cRNAs.

We used One-Color Microarray-Based Gene Expression Analysis (Quick Amp Labeling) v5.5 and according to the manufacturer’s instructions.

Data processing |
The intensities were measured using Agilent Feature Extraction Software 9.5 (Agilent Technologies). A global normalization was performed for the inter-chip normalization. In order to define the expression level of the gene/probe, the signal intensity of each gene on a chip was divided by the median value of the signal intensities of the entire genes on the chip.
