Total RNA from the pooled prostate tissue from group 1 (1500 ng) and groups 2-4 (300 ng each) was reverse transcribed into cDNA with MMLV-RT and oligo dT-promoter primer, and the resulting cDNA was used for the synthesis of fluorescently labeled cRNA sample probes with the Agilent Quick Amp Labeling Kit, following manufacturer's protocol.

Hybridization protocol |
Blocking Agent and Fragmentation Buffer (Agilent Gene Expression Hybridization kit) were added to the samples, and incubated at 60 C for 30min. GE Hybridization Buffer HI-RPM was added, and samples were applied to microarrays enclosed in Agilent 4x44K gasket slides. Slides were incubated at 65 C for 1 day, and washed sequential with Agilent Gene Expression Wash Pack, following manufacturer's protocol.

Scanned on an Agilent G2505A scanner. Images were quantified using Agilent Feature Extraction Software (v 9.5.1.1).

Description |
Biological replicate 1 of 3. Partial bladder outlet obstruction. Treated twice a day with 18 mg/kg Eviprostat for five days.

Data processing |
Agilent Feature Extraction Software (v 9.5.1.1) was used for background subtraction and LOWESS normalization.
