Growth protocol The rhesus monkey embryonic cells derived from flushed preimplantation embryohas been characterize for its pluripotency including its potential differentiation to neural stages. All reagents in the cell culture experiments were used from Invitrogen corporation. ESC cell from passage 13 were collected as (ESC) and propagated on MEF layer with DMEM Eagle's media.(ESC stage). after 6-8 days 1-1.5 mm embroid bodies were collected as (EB). EBs werew then cultured in DMEM/F12 mudium with N2 supplement for 7-9days.After 9days 0..25-1.25 mm speres of N1 collected(N1stage). N1 further grow in the medium MDEM/F12/N2 suppliment and added FGF4 for expansion of Nestin positive cell and collected after 7-9days(N2 stage). The neurospheres of stage N2 further extended in neuronal maturation phase with in GFR reduced well in DMEM/F12 medium without FGF4.
