Treatment protocol Ovariectomized cows (n=12) were randomly divided into four groups; the control group (n=3) received saline treatment at Day 0 (beginning of the experiment), E2 groups (n=3) received saline and 1 mg estradiol benzoate (EB) at Day 0 and Day 5, respectively, two CIDR were inserted to the uterus of animals of the P4 group (n=3) from Day 0 to Day 6, and the E2+P4 group (n=3) received two CIDR at Day 0 and injection of 1 mg EB immediately after removal of the CIDR at Day 6. Blood samples (5 ml) in control and P4 groups were collected at Day 5 after saline or CIDR treatment, and in E2 and E2+P4 groups blood samples were collected after EB treatment. All animals were slaughtered 2 days after blood sampling, and endometrium samples were collected as described in Bauersachs et al J Mol Endocrinol 2005). The tissue samples were snap-frozen and stored at -80°C until analysis.
Growth protocol The cows (n=12) were ovariectomized, maintained in a free-stall barn throughout the experimental period, and had free access to food and water.
Extracted molecule total RNA
Extraction protocol deep frozen tissue was transferred to 5 ml RNAlater-ICE (Ambion, Austin, TX, USA) and allowed to impregnate at -20°C during two days for optimal RNA protection. Total RNA was extracted from bovine mammary gland tissue with the Trizol method according to the manufacturers protocol
Label biotin
Label protocol total RNA was transcribed and labeled with Ambion Kit MessageAmp II Biotin Enhanced according to the manufacturers protocol
 
Hybridization protocol fragmented cRNA was hybridized to Affymetrix GeneChip Bovine Genome Arrays and washed and stained with Affymetrix Wash&Stain Kit on an Affymetrix Fluidics Station 450
