Extraction protocol Schwann cell samples were randomly selected from control groups (n = 3) and diabetic groups (n = 3), rinsed with PBS twice and total RNA was isolated by using miRNeasy Mini Kit (Qiagen, Germany) according to the manufacturer’s instructions.
RNA libraries were prepared for sequencing using standard Illumina protocols

Data processing Illumina Xten software used for basecalling.
Transcriptome sequencing uses high-throughput sequencing technology to sequence the cDNA after total RNA reverse transcription, and obtains most transcripts of samples in this state.
Sequenced reads were trimmed for adaptor sequence, and masked for low-complexity or low-quality sequence, then mapped to whole genome and FPKM were calculated using StringTie(Pertea et al., 2016)
