Treatment protocol 8-12 weeks old female YAPfl/fl mice were administered AAV8.TBG.PI.Cre.rBG (2.5 E11 viral particles per mouse, i.p.) to obtain hepatocyte specific YAP KO mice. AAV8.TBG.PI.eGFP.WPRE.bGH8 (2.5 E11 viral particles per mouse, i.p.) administered to YAPfl/fl mice served as wild-type (WT) control. Single dose of TCPOBOP (3 mg/kg; prepared in corn oil) was administered by oral gavage 10 days after AAV8 injection. Mice were sacrificed at various time points (0, 1 and 2 days) after TCPOBOP administration. Day 0 animals did not receive any treatment except AAV8 administration.
Growth protocol Following hepatocyte-specific deletion of YAP using AAV8-TBG-CRE, mice were treated with chemical mitogen TCPOBOP and liver tissues harvested at 0, 1, and 2 days after TCPOBOP treatment.
Extracted molecule total RNA
Extraction protocol Trizol extraction of total RNA was performed according to the manufacturer's instructions.
Label biotin
Label protocol Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 6 ug total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).
Hybridization protocol Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on GeneChip mouse Genome Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.
Scan protocol GeneChips were scanned using the Hewlett-Packard GeneArray Scanner G2500A.
Description Gene expression data at Day 2 post-TCPOBOP in YAP-KO mice
Data processing The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings and global scaling as normalization method. The trimmed mean target intensity of each array was arbitrarily set to 100.
