Treatment protocol Embryo were dechorionated with proteinase K and homogenized by Buffer RLT (Rneasy@ Mini Kit, Qiagen).
Growth protocol Embryos were obtained from crossing of stac heterozygous and classified by phenotype and EGFP expression. These embryos were raised in holfreter water and collected at 24hpf and 30hpf.
Extracted molecule total RNA
Extraction protocol Total RNA was isolated with Rneasy@ Mini Kit according to the protocol "Purification of total RNA from animal tissues" on Rneasy@ Mini Handbook (Quagen).
Label biotin
Label protocol Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 8ug total RNA, and labeling was carried out at 37C for 16hr. Biotinylated cRNA was purifed by QIAGEN RNeasy Total RNA Isolation kit.
 
Hybridization protocol Following fragmentation,1 ug of cRNA were hybridized for 16 hr at 45C on Affymetrix GeneChip® Zebrafish Genome Array with Hybridization Oven 640: Affymetrix, 800139 (220V). GeneChips were washed and stained in the Fluidics Station 450: Affymetrix, P/N 00-0079
Scan protocol GeneChips were scanned using the GeneChip® Scanner 3000: Affymetrix, P/N 00-00212
Description Gene expression data from mutant (stac) embryos
Data processing The data were analyzed with Microarray Suite 5.0
