Treatment protocol |
Common progenitor cells were cultured with M-CSF alone for macrophages and M-CSF plus RANKL for osteoclasts, respectively. After six days of cultivation, cells were lysed in RLT solution (RNeasy, QIAGEN).

Growth protocol |
Bone marrow cells were isolated from 8 weeks old mouse and cultured in the presence of M-CSF for three days. Then M-CSF-dependent adherent cells were harvested as macrophage and osteoclast common progenitor cells.

RLT extraction of total RNA was performed according to the manufacturer's instructions (Qiagen).

Biotinylated cRNA were prepared according to the standard Affymetrix protocol, one-cycle, from 1 ug total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).

Hybridization protocol |
Following fragmentation, 15 ug of cRNA were hybridized for 16 hr at 45C on GeneChip®Mouse Genome 430 2.0 Array. GeneChip® Arrays were washed and stained in the Affymetrix Fluidics Station 400.

Scan protocol |
GeneChips® Arrays were scanned using the GeneChip® Scanner 3000 7G.
