| Treatment protocol |
To inactivate Myb, estrogen was withdrawn by washing cells with PBS thrice and culturing them in IMDM supplemented with 10% FBS, 60 FDU/mL of GM-CSF and 100 nM of β-estradiol antagonist ICI-182780. ChIP samples were prepared from Myb-on cells (grown in the presence of estrogen) or Myb-off cells (estrogen withdrawn for 6 hr).

ERMYB cells were maintained in IMDM supplemented with 10% fetal bovine serum, 60 FDU/mL of GM-CSF and 1 μM of β-estradiol.

Lysates were clarified from sonicated nuclei and Myb-DNA complexes were isolated with antibody. Libraries were prepared according to Illumina's instructions accompanying the DNA prep Sample Kit (Part# 0801-0303). The purified DNA was captured on an Illumina flow cell for cluster generation. Libraries were sequenced on the Genome Analyzer II following the manufacturer's protocols.

Alignment: Sequence reads were obtained and mapped to the mouse genome (USCS build mm9) using the Illumina Genome Analyzer Pipeline v.1.3. Reads with ambiguos aligmnets (2 or more hits) were excluded from further analyses.
