Treatment protocol |
KRAS G12V and p53 R175H lentiviruses and PIK3CA E545K retroviruses were used to establish the BEAS2B stable cell lines. Cell lines were infected with viral-containing supernatants for a period of 4-8 hours, and then the infected cultures were selected by puromycin (PIK3CA vectors), GFP-sorting (KRASG12V) or blasticidin (p53R175H).

Growth protocol |
BEAS-2B were propagated in RPMI-1640 media containing 1x penicillin/streptomycin, 4mM L-glutamine, 10% fetal bovine serum in 10 cm gelatin-coated petri-dishes.

Extraction protocol |
RNA from treated cell lines was extracted using Absolutely RNA Mini-prep kits (Agilent). RNA libraries were prepared for sequencing using BGI protocols.

Data processing |
Sequencing and base-calling were performed by BGI (Beijing Genomics Institute). Sequencing reads were trimmed and filtered using Trimmomatic (V0.39) to remove adapters and low quality reads. Reads were mapped to Ensembl GRCh38 transcripts annotation (release 98), using RSEM (v1.3.2) and STAR (2.6.1d) with default settings.
