Homogenization and lysis of cells with Lysing Matrix D (Qbiogene) in Trizol LS (Invitrogen) with FastPrep120 (Qbiogene) instrument. Purification with Qiagen RNeasy Mini kit. All performed according to the manufacturers' instructions.

5 ug total RNA was processed according to the Affymetrix recommended protocol for Eukaryotic samples, Affymetrix technical manual, revison 5.

Hybridization protocol |
Fragmented, biotinylated cRNA was hybridized to Plasmodium/Anoptheles GeneChips for 16hours at 45oC and 60rpm. Affymetrix recommended signal amplifcation protocol for washing/staining of Eukaryotic targets was performed in a FS450 automated fluidics station.

GeneChips were scanned in an Affymetrix GCS3000 scanner with autoloader.
