Extraction protocol TRIzol was used for the RNA extraction, manufacturer's instructions were followed

Label protocol rRNA reduced samples were amplified and labelled using Genechip WT cDNA Synthesis/Amplification Kit and GeneChip WT Terminal Labeling Kit (Affymetrix, Santa Clara, CA)
 
Hybridization protocol Samples were processed using the Affymetrix hybridization, wash and stain kits. 200µl of the hybridization mixture was hybridized onto each Mouse Exon 1.0 st array. Hybridized at 45°C with 60rpm rotation for 17 hours in Affymetrix hybridization oven 640
Scan protocol Affymetrix Genechip scanner 3000 7G
Description Mta1 is re-expressed in the knockout MEFs
Data processing Data were processed using Genespring GX 10.0.2 (Affymetrix Technologies,Inc) . The raw data was summerized using RMA16 algorithm which is inbuilt in Genespring GX,followed by the log transformation, baseline transformation and finally quantile normalization.
