Description B10.C-H7b(47N)/SN female mice were immunized by i.p. co-injection of C57BL/10Snj male and B10.C-H7b(47N)/SN male spleen cells. Fourteen days later, single-cell suspensions were prepared from the spleen of immunized mice. CD8+H7aTet+, CD8+HYTet+ and CD8+Tet- cells were sorted using a FACSVantage SE with Diva option (BD Biosciences, San Jose). Purity of sorted cell populations was always ≥ 98%. Tet+ populations containing between 8-300 thousand cells and Tet- population containing 2-3 million cells were obtained for each mouse and processed separately.
