Hepatocytes were isolated after liver perfusion with EBSS, 0.5mM EGTA and EBSS, 10mM Hepes, collagenase, trypsin inhibitor, snap-frozen and stored at -80°C until RNA extraction.

Adult mice were treated with drinking water containing 4mg/ml doxycycline/ 10% sucrose for 8 days to induce miR-143 overexpression in miR-143DOX mice.

Following fragmentation, cDNAs were hybridized for 17 hr at 45°C on Affymetrix Mouse Gene 1.0 ST Arrays. GeneChips were washed and stained in the GeneChip Fluidics Station 450 (Affymetrix).
