Title |
Sham control, biological rep3 |
Sample type |
RNA |
Source name |
Saline-injected sham control, biological rep3
Organism |
Rattus norvegicus |
Characteristics |
strain: Sprague-Dawley age: 12 ~ 14 weeks tissue: articular cartilage agent: control time: n/a
Treatment protocol |
Monoiodoacetate (MIA, 2 mg) dissolved in 50 ul of saline was intra-articularly injected into a knee of hinder limbs. Articular cartilage in the distal end of femur was exercised after 5, 9 or 21 days post-injection. Saline injected rats were used as sham controls.
Extracted molecule |
total RNA |
Extraction protocol |
Trizol extraction of total RNA was performed according to the manufacturer's instructions.
Label |
biotin
Label protocol |
Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 300 ng of total RNA according to Affymetrix's protocols.
Hybridization protocol |
2ug of labeled and fragmented DNA was hybridized at 45C for 17 hours at 60rpm. GeneChips were washed and stained on the Affymetrix Fluidics Station 450 according to the manufacturers protocol.
Scan protocol |
GeneChips were scanned with the Affymetrix GeneChip Scanner 3000 7G.
Description |
JN-3.CEL Gene expression data from articular cartilage in the distal end of femur from sham control
Data processing |
The data were analyzed by Partek Genomics Suite version 17 using GC-RMA normalization.
