Mouse ES cells were maintained on irradiated embryonic fibroblast feeder cells and cultured in ES medium (DMEM supplemented with 2 mM L-glutamine, 1 mM sodium pyruvate, 0.1 mM 2-mercaptoethanol, 0.1 mM nonessential amino acids, 50 u/ml Penicillin/Streptomycin, 15% ES cell-qualified FBS, and 1000 u/ml LIF (Millipore)). Cells were passaged twice on 0.1% gelatin-coated dishes to remove feeder cells and then cultured in ES medium on gelatin-coated dishes.
