Treatment protocol At ~44 weeks of age, the animals (n=9-10) were gavaged twice daily for 28 days with the specified doses of RAD001 and/or BEZ235. The mice were starved for three hours prior to sacrifice and received their last dose of RAD001 at the time starvation or BEZ235 1 hour before sacrifice.
Growth protocol C57Bl/6 mice were obtained from Jackson laboratories. At two weeks of age, male mice were injected intraperitoneally with 50 mg/kg BW of diethylnitrosamine (DEN) (Sigma).
Extracted molecule total RNA
Extraction protocol Total RNA from all the samples was purified using the Qiagen RNeasy purification kit following the manufacturer’s protocol.
Label biotin
Label protocol The integrity of the RNA was measured using a Bioanalyser and RIN values > 7-8 were considered adequate to proceed further with total RNA transcription into cRNA using the Illumina Total Prep RNA amplification kit (Ambion) following the manufacturer’s protocol.
 
Hybridization protocol Biotinylated cRNA was hybridized to Illumina MouseRef8v2 bead chips.
Scan protocol Gene expression matrices were scanned using Illumina iSCAN Control Software.
Description 30 mg/Kg BW
Data processing Data was quantile normalized with background subtraction using Illumina BeadStudio.
