Title |
AC_activated control |
Sample type |
RNA |
Source name |
4h culture of activated AC without compound treatment
Organism |
Mus musculus |
Characteristics |
cell line: Rosa25/OG2 cell type: Neural stem cell (NSC)-derived astrocytes (AC) activated with: 1 µg/ml LPS and 10 ng/ml IFN-γ
Treatment protocol |
Medium was switched to DK10 and cells were activated with 1 µg/ml LPS and 10 ng/ml IFN-γ, followed by treatment with the indicated hit compound for 4 h. Cells were harvested directly on the culture dish.
Growth protocol |
Astrocytes were differentiated from NSCs on gelatine in medium containing 10% FCS for 4 days and replated at a density of 1.5 x 1,000,000 cells/cm2 on Matrigel-coated cell culture dishes and matured for 14 days before treatment.

Label protocol |
Ambion TotalPrep RNA Amplification Kit. RNA in put: 400ng, IVT: 12h
Hybridization protocol |
According to manufacturer's instructions. Hybridization time: 18h
Scan protocol |
Illumina iScan system using default settings
Data processing |
Cubic Spline normalization with background subtraction in Illumina GenomeStudio V2010.3 with the module GeneExpression 1.8.0. Signal intensities below 10 were arbitrarily trimmed to the value of 10.
