Title |
COPD [275177_miRNA] |
Sample type |
RNA |
Source name |
Lung tissue from smoker with COPD
Organism |
Homo sapiens |
Characteristics |
diagnosis: COPD fvc-group: 2 gold stage: 2 age: 64 gender (1=male, 2=female): 2 pkyrs: 52
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA was isolated from frozen tissue samples using a freeze fracture protocol, followed by Trizol extraction and precipitation at -20ºC overnight to increase the yield of small RNAs. RNA integrity was determined by capillary electrophoresis on an Agilent 2100 Bioanalyzer. Only RNA with an integrity number $7 was used in the profiling studies.
Label |
Cy5
Label protocol |
miRNAs were labeled according to the manufacturer's instruction (Exiqon, Woburn, Massachusetts, USA)
Hybridization protocol |
The labelled miRNA probes were hybridized to the miRCURY LNA arrays v11.0 for 16h at 56ºC
Scan protocol |
Hybridized slides were washed and scanned with ScanArray Express (Perkin Elmer)
Description |
COPD
Data processing |
miRNA expression data was consolidated by taking median value across replicated probes. Then, it was normalized by quantile method, and transformed in log2-scale
