24 d old Il2-/- mice were injected intraperitoneally with 50 ng/g body weight of recombinant human IL-2 in PBS every 8 h for 24 h (total of four injections). After 24 h, lymph node and spleen suspensions were subjected to a combination of MACS and FACS sorting for RNA isolation as described in Nat Immunol. 2005 Nov;6(11):1142-51.
