Cloning and Expression

Byk et al. (1998) identified human ESTs representing ULIP4 that were isolated from a brain cDNA library. Using one of these sequences as probe, they cloned mouse Ulip4 from a whole newborn mouse brain cDNA library. The deduced 572-amino acid protein contains several phosphorylation consensus sites conserved in other Ulip proteins. Northern blot analysis of several mouse tissues detected Ulip4 expression only in brain. RNA dot blot analysis of developing whole rat brain showed an increase in Ulip4 expression between embryonic and perinatal stages, and slightly lower expression in adult animals.

Honnorat et al. (1999) found that autoantibodies identified in patients with paraneoplastic neurologic diseases recognized a 66-kD protein representing several posttranslationally modified forms of ULIP4. By EST database analysis, they identified a clone encoding a deduced 572-amino acid protein that is 92% identical to mouse Ulip4.

By in situ hybridization, Quach et al. (2000) demonstrated that mouse Ulip4 is expressed mainly in the dentate gyrus of hippocampus, the granular layers of cerebellum, and the inferior olive of the pons.


Gene Structure

Quach et al. (2000) determined that the mouse Ulip4 gene contains 14 exons and spans more than 20 kb.


Mapping

By FISH, Honnorat et al. (1999) mapped the ULIP4 gene to chromosome 10q25.2-q26.

By interspecific backcross analysis, Quach et al. (2000) mapped the mouse Ulip4 gene to the distal end of chromosome 7 in a region that shows homology of synteny to human chromosome 10q26.
