Members of the FASTKD family of proteins, like FASTKD4, are predicted to be mitochondrial proteins that sense mitochondrial stress and modulate protein translation to promote survival of cells exposed to adverse conditions (Simarro et al., 2010).
By screening a hepatoma cDNA expression library for clones that could block mating pheromone-induced G1 arrest in yeast, Edwards et al. (1997) cloned TBRG4, which they called CPR2. The deduced protein contains multiple putative leucine zipper domains characteristic of transcription factors.
By sequencing clones obtained from a size-fractionated human brain cDNA library, Nagase et al. (1999) cloned TBRG4, which they designated KIAA0948. The transcript contains several repetitive elements in the 3-prime untranslated region. The deduced 521-amino acid protein shares significant similarity with rat Cpr2. RT-PCR ELISA detected moderate expression in ovary, and little to no expression in all other tissues and specific brain regions examined.
Simarro et al. (2010) reported that the deduced 631-amino acid human FASTKD4 protein has a mitochondrial targeting signal in the N terminus and 2 FAST domains and a RAP domain, which is predicted to bind RNA, in the C terminus. Qualitative RT-PCR detected variable FASTKD4 expression in all human and mouse tissues examined. In human tissues, highest FASTKD4 expression was detected in skeletal muscle. In mouse tissues, highest Fastkd4 expression was detected in brown adipose tissue, heart, testis, and skeletal muscle. Fluorescence-tagged FASTKD4 colocalized with a mitochondrial marker in transfected cells.
