Cloning and Expression

By screening for long cDNA clones expressed in an immature myeloid cell line, Nagase et al. (1996) cloned TMEM131, which they called KIAA0257. The deduced 1,805-amino acid protein contains a transmembrane domain and an ATP synthase motif. Northern blot analysis revealed ubiquitous expression, with highest expression in skeletal muscle.

Using mRNA differential display, Tscharke et al. (2000) identified Tmem131, which they called Cc28, based on its expression in ganglia of immunocompetent mice infected with herpes simplex virus (HSV) but not in ganglia of uninfected or HSV-infected mice depleted of Cd8 (see 186910) T cells. The full-length Cc28 protein contains 1,825 amino acids and shares 93% amino acid identity with human KIAA0257. Cc28 contains 2 potential transmembrane domains. Northern blot analysis revealed variable expression of a 6.5-kb transcript in all mouse tissues and mouse, rat, and monkey cell lines examined; expression was not detected in human HepG2 cells.


Mapping

By radiation hybrid analysis, Nagase et al. (1996) mapped the KIAA0257 gene to chromosome 2.

Gross (2014) mapped the TMEM131 gene to chromosome 2q11.2 based on an alignment of the TMEM131 sequence (GenBank BC036841) with the genomic sequence (GRCh37).
