Study: mRNA-Seq of brain tissue from honeybee (Apis mellifera) drones (male), workers (female) and queens (female).
show Abstracthide Abstract
Transcriptional profiling of brain tissue of was performed by RNA-SEQ in workers, queens and males. The sex and caste specific expressed and spliced genes were examined comparing drone, worker and queen honeybees. Overall design: Examination of mRNA profiles in heads of male, worker and queen honeybees pupae were compared.

Layout: SINGLE
Construction protocol: Total RNA was isolated from single brains using Trizol method. 50 µl Trizol were added to a single brain and the tissue was pestled. After incubation for 5 min at room temperature 10 µl chloroform were added. The solution was mixed for 30 sec and centrifuged for 15 min on 13,000 rpm. The upper phase was transferred into a new tube and 20 µl isopropanol were added and mixed. The solution was incubated for 10 min at room temperature and then centrifuged for 10 min at 13,000 rpm and 4°C. The pellet was dired for 5 min at room temperature and then diluted in 10 µl RNAse free water.
