Design: Samples were extracted using Qiagen DNeasy extraction kits. DNA was sheared to 400-600 bp using a QSoncia Q800R sonicator. Genomic libraries were prepared using Kapa Hyper Prep Kits (1/4 volume reactions) and iTru, 8bp dual-indexing adapters (Glenn et al. 2019). Samples were pooled into pools of 10 samples and enriched for ultraconserved elements (UCEs) using an ant-customized RNA bait set (Arbor Biociences; ant-specific hym-v2) that includes 9,898 baits (120 mer) targeting 2,524 UCE loci shared across Hymenoptera and a set of legacy markers (Branstetter et al. 2017). Enriched pools were pooled together (up to 104 individual samples) and shipped to the University of Utah genomics core facility for sequencing on an Illumina HiSeq 2500 instrument (2x125 bp v4 chemistry).

Study: Phylogenomic species delimitation and bird guide taxonomy in the Neotropical ant genus Rasopone (Hymenoptera: Formicidae)

Rasopone Schmidt & Shattuck is a poorly known lineage of ants that live in Neotropical forests. Incorporating results from ultraconserved element (UCE) phylogenomics and DNA barcodes, we revised the genus, providing a new morphological diagnosis and a species-level treatment. Our study provides foundational descriptive taxonomy that will facilitate research on the biogeography, ecology, and evolutionary history of ants in the Middle American corridor, while also demonstrating the value of integrating phylogenomic data into species delimitation.
