Study: RNA-Seq analysis between the wild-type and AmbldC mutant strains

The transcriptomes of the rare actinomycete Actinoplanes missouriensis wild-type and AmbldC deletion mutant strains were analyzed by the RNA sequencing technique with the Illumina HiSeq2500 sequencer. The total RNA samples were extracted from the mixtures of mycelia and sporangia cultivated on the HAT agar at 30 degrees for 3 days. AmBldC is the transcriptional regulator that is actively produced in the early stage of sporangium foramtion and is required for normal sporangium dehiscence in A. missouriensis. In this study, the transcriptomes of the wild-type and AmbldC mutant strains were compared to define the AmBldC regulon to characterize the molecular mechanisms of sporangium formation and dehiscence in A. missouriensis.
