We have determined that chemical FoxO1 inhibition can effectively mimic many aspects of beta cell dedifferentiation in vitro and that this can be countered by loperamide treatment. Overall design: RNAseq results of murine beta cell line, Min6, treated for 48h with DMSO, AS1842856 (FoxO1 inhibitor), loperamide and in combination.

Construction protocol: RNA was isolated using the RNeasy Micro Kit (Qiagen), including DNase treatment, following the manufacturer's instructions. RNA-seq libraries were prepared with the TruSeq Stranded mRNA LT sample preparation kit (Illumina) using Sciclone and Zephyr liquid handling robotics (PerkinElmer). Library amounts were quantified using the Qubit 2.0 Fluorometric Quantitation system (Life Technologies) and the size distribution was assessed using the Experion Automated Electrophoresis System (Bio-Rad). For sequencing, libraries were pooled, diluted and sequenced on an Illumina HiSeq 4000 instrument using 50 bp single-read chemistry.
