Design: Cell suspension from digested leg bones were pooled together with flushed BM fraction after CD45+ leukocyte depletion. The RNA was extracted using RNeasy Mini Kit (QIAGEN) and DNA was removed using RNase-Free DNase Set (QIAGEN). RNA-seq was performed by BGI Genomics (Hong Kong). Library selection: poly-A selection method.

To define mechanisms by which SBDS deficiency in bone marrow (BM) niches impairs donor hematopoietic stem cell engraftment after transplantation, we performed RNA-seq on the BM stromal cells from Sbds-deficient mice versus control mice at 24 hours after myeloablative total body irradiation (TBI) and at baseline.
