The goals of this study are to investigate the molecular mechanism and biomarker by DNA methylation profiles in patients with metabolic syndrome compared to normal. Overall design: In this study, we performed Infinium MethylationEPIC BeadChIP and RNA-seq with same patients to explore a molecular targets for the prediction of metabolic syndrome. A total of 20 participants, including 11 patietns with metabolic syndrome and 9 normal were enrolled in our study.

Construction protocol: Total RNA was isolated from PBMC cell using TRIzol® RNA Isolation Reagents (Life technologies, Carlsbad, CA) based method. The quantity and quality of the RNA were evaluated using Agilent Nono Kit and assessing the RNA integrity number (RIN >7). One mg of total RNA was processed for preparing mRNA sequencing library using MGIEasy RNA Directional Library Prep Kit (MGI) according to manufacturer's instruction. RNA libraries were prepared for sequencing using standard MGI protocols
