Study: Transcriptome dynamics of hematopoietic stem cell formation revealed using a combinatorial Runx1/Ly6a reporter system
show Abstracthide Abstract
Single cell RNA-Seq time-course analysis revealed that as pre-HSCs mature into fetal liver-stage HSCs they show signs of interferon exposure, multi-lineage differentiation gene expression signatures, and prolonged cell cycle reminiscent of quiescent adult HSCs. Overall design: Investigation of transcriptome dynamics of hematopoietic stem cell formation using a combinatorial Runx1/Ly6a reporter system
Sample: single cellf from YS [LIB019717_GEN00050537_S84_R1]

Organism: Mus musculus
Library:
Instrument: Illumina HiSeq 2500
Strategy: RNA-Seq
Source: TRANSCRIPTOMIC
Selection: cDNA
Layout: PAIRED
Construction protocol: Embryos were harvested from the uterus and dissected in cold dissecting buffer (Dulbecco's phosphate buffered saline with 10% fetal bovine serum and antibiotics). Dissected tissues were dissociated in 0.125% collagenase type I (Sigma-Aldrich) and 1 U Dispase (Stemcell Technologies) at 37°C for 30 min. The samples were reduced to single cell suspension by mechanical trituration, washed with dissecting media, and passed through 40mm cell strainer. FACS was used to enrich single cell population of interest Cells were lysed and single cell libraries prepared by the C1 System using the SMARTer Ultra Low RNA Kit (Clontech) and protocols provided by Fluidigm.
