1 ILLUMINA (Illumina HiSeq 2500) run: 599.6M spots, 121.1G bases, 52.7Gb downloads

Design: We performed DNA-seq PCR-free as previously described (Blecher-Gonen, Barnett-Itzhaki, Jaitin, Amann-Zalcenstein, Lara-Astiaso, and Amit 2013), with the following modifications: Two samples of ~1ug of genomic Rat liver DNA were sheared to a peak of 291 and 286bp, using the Covaris S220 sonicator (Covaris, Inc., Woburn, MA, USA). We performed end repair in 80ul reaction at 20oC for 30min. Following cleanup with Agencourt AmPURE XP beads (Beckman Coulter, Inc., Indianapolis, IN, USA) at a ratio of 0.75X Beads/DNA volume, we added bases to both 3’ ends. We ligated adapters (Bioo Scientific Cat# 514112) in a final concentration of 1.1uM following a SPRI beads cleanup at a ratio of 0.75x beads/DNA volume. Sample preparation was without the PCR step. Libraries quantitation was in 50 Single Read HigSeq runs.

We sequenced (NGS) the genomes of the Cohen Diabetic sensitive (CDs/y) and the Cohen Diabetic resistant (CDr/y) strains, a fully inbred model of diet-induced type 2 diabetes, in order to detect sequence variations of potential relevance to the diabetic phenotype.
