Design: DNA was randomly fragmented by Covaris. Fragmented DNAs were tested by Agilent 2100 and purified by the Agencourt AMPure XP kit. The selected fragments were through end-repair, 3'adenylated, adapters-ligation and purified by the Agencourt AMPure XP kit. These fragments were amplified with KAPA HiFi HotStart DNA Polymerase and the PCR products were purified with the Agencourt AMPure XP kit. The double stranded PCR products were heat denatured and circularized by the splint oligo sequence. The single strand circle DNA (ssCir DNA) were formatted as the final library and qualified by QC
