Platelet derived growth factor effect in the presence of Src family kinase inhibitors (MOE430A)

Analysis of NIH3T3 fibroblasts treated with platelet-derived growth factor (PDGF) and one of several Src family kinase (SFK) small molecule inhibitors. Inhibitors of SFKs, MEK1/2, and phosphotidylinositol 3-kinase used. Results provide insight into the role of SFKs in PDGF signaling.

PDGF-induced genes regulated by the PI3K and MEK/ERK signaling pathways

(Submitter supplied) Human T98G glioblastoma cells were stimulated with platelet-derived growth factor (PDGF) and analyzed by DNA microarrays, which identified 74 immediate-early gene transcripts. Cells were then treated with inhibitors to identify subsets of genes that are targets of the phosphatidylinositol 3-kinase (PI3K) and MEK/ERK signaling pathways. Four groups of PDGF-induced genes were defined: independent of PI3K and MEK/ERK signaling, dependent on PI3K signaling, dependent on MEK/ERK signaling, and dependent on both pathways. more...

Combinatorial treatments targeting MAPK and PI3K/mTOR pathways in metastatic melanoma

(Submitter supplied) Therapeutic targeting of BRAFV600E has shown a significant impact on progression-free and overall survival in advanced melanoma, but only a fraction of patients benefit from these treatments, suggesting that additional signaling pathways involved in melanoma growth/survival need to be identified. In fact MAPK and PI3K/mTOR signaling pathways are constituively activated in most cancers, including melanoma, to sustain the melanoma growth/survival. more...
