Exposure to polybrominated biphenyl (PBB) associates with DNA methylation differences across the genome

(Submitter supplied) Polybrominated biphenyl (PBB) is an endocrine-disrupting compound which was accidently added to the food supply in Michigan in the 1970’s. Highly exposed individuals and their children have numerous health problems, though the underlying mechanism behind these health problems remains unknown. Other endocrine-disrupting compounds have been linked to epigenetic differences, but no epigenetic studies have been done for PBB. more...

(Submitter supplied) Genome wide DNA methylation profiling of monocytes from insulin sensitive and insulin resistant HIV-infected patients. The Illumina Infinium Human Methylation 450k Beadchip was used to obtain DNA methylation profiling across 450,000+ CpGs. Samples included monocytes from 37 HIV-infected individuals, of which 14 were insulin resistant and 23 were insulin sensitive, monocytes from 9 HIV-seronegative individuals, of which 4 were insulin sensitive and 5 insulin resistant, and FACS monocyte subsets (classical, intermediate, and non-classical monocytes) from two healthy donors.

DNA methylation profiles of human blood samples from Hispanic subjects with Parkinson's disease

(Submitter supplied) Human DNA methylation Beadchip v1.2 was used to profile n=84 whole blood samples from Hispanics living in the USA. The main goal of the study was to relate Parkinson's disease status to measures of epigenetic age acceleration based on DNA methylation data. Here we focus on Hispanic subjects. To measure DNA methylation age, we used the epigenetic clock software described in Horvath S (n=2013) DNA methylation age of human tissues and cell types. more...

Aging-associated DNA methylation changes in middle-aged individuals: The Young Finns Study

(Submitter supplied) Background Chronological aging-associated changes in the human DNA methylome are studied by multiple epigenome-wide association studies (EWAS); however, the aging-associated DNAmet changes identified among different age groups and tissues vary and especially the rates of aging-associated alterations in the epigenome during adulthood remain unclear. Here, we further explore and characterize CpG-sites where DNA methylation levels alter at a constant rate during adulthood and are also independent of blood cell type heterogeneity. more...

Mapping of variable DNA methylation across multiple cell types defines a dynamic regulatory landscape of the human genome

(Submitter supplied) DNA methylation is an important epigenetic modification involved in many biological processes and diseases. Many studies have mapped DNA methylation changes associated with embryogenesis, cell differentiation and cancer at a genome-wide scale. Our understanding of genome-wide DNA methylation changes in a developmental or disease-related context has been steadily growing. However, the investigation of which CpGs are variably methylated in different normal cell or tissue types is still limited. more...

Breast cancer risk factors are associated with DNA methylation in non-diseased breast tissue independent of cell type

(Submitter supplied) The underlying biology through which established breast cancer risk factors contribute to disease risk is not well characterized. One key risk factor for breast cancer is age, and age-related DNA methylation alterations may contribute to increased risk of disease. Here we assessed normal breast tissues and tested the relation of DNA methylation with known breast cancer risk factors. Cancer-free women donated breast tissue biopsy specimens through the Susan G. more...
