Gamma-tocotrienol effect on breast cancer MCF-7 cells

Analysis of MCF-7 cells treated with gamma-tocotrienol (γ-T3). Tocotrienols (T3s) inhibit the growth of various cancer cell lines without affecting normal cells. Results provide insight into the molecular mechanisms of action of T3s on cell death and other growth inhibitory pathways.

Expression data of MCF-7 cells treated with gamma tocotrienol (g-T3)

(Submitter supplied) Gamma tocotrienol induces apoptosis in breast cancer cells however, the molecular mechanisms are not completely understood. We used microarrays to detail the global programme of gene expression underlying the effects of gamma tocotrienols on MCF-7 cells and identified distinct classes of up-regulated genes during this process.

RNA sequencing of MDA-MB231 and U2OS cancer cell lines exposed to the alkylating agent methyl methanesufonate (MMS) and classical chemotherapeutics

(Submitter supplied) Understanding the mechanisms by which cells respond to chemotherapeutics is key to identifying means to improve therapy effiicacy while reducing systemic toxicity of these widely used classes of drugs. While determining the role of NRF2-GSH and ER stress in cells exposed to alkylating compounds such as methyl-methanesulfonate (MMS), we asked if these pathways could also be a general cell damage response relevant to other clinically used chemotherapeutics or if it is an alkylation specific response. more...

Effect of actein on the growth of the MDA-MB-453 breast cancer cell lines as a function of time and concentration.

(Submitter supplied) Previous studies indicate that the triterpene glycoside actein from the herb black cohosh inhibits growth of human breast cancer cells. This study seeks to identify genes altered in human breast cancer cells by treatment with actein, using gene expression analysis. We treated MDA-MB-453 human breast cancer cells with actein at 2 doses, 20 or 40 μg/mL, for 6 or 24 h. We identified 5 genes that were activated after each of the treatments that are known to play a role in cellular responses to diverse stresses, including the DNA damage and unfolded protein responses. more...

Actein effect on breast cancer cell line: dose response and time course

Analysis of MDB-MB-453 breast cancer cells treated with 20 or 40 ug/ml actein for 6 or 24 hours. Actein is a triterpene glycoside from the herb black cohosh and inhibits the growth of cancer cells in vitro. Results provide insight into the molecular basis of this inhibitory effect.

RHBDD2 silencing and gene expression profiling of breast cancer cells

(Submitter supplied) In previous studies, we identified a distantly related rhomboid homologue gene known as RHBDD2 (Rhomboid domain containing 2) to be markedly overexpressed in the advanced stages of the breast and colorectal cancer diseases. In order to identify RHBDD2 modulated pathways, we analyzed two breast cancer cell lines (MCF7 and T47D) from control and RHBDD2-siRNA transient gene silencing followed by gene expression profiling analysis using the whole genome Toray 3D-GeneTM Human Oligo Chip. more...

17beta-estradiol (E2)-regulated gene expression across time in estrogen-dependent and estrogen deprivation-resistant MCF-7-derived breast cancer cell lines.

(Submitter supplied) A series of MCF-7 variants were previously developed that are either estrogen-dependent for growth (MCF-7:WS8 cells), or resistant to estrogen deprivation and refractory (MCF-7:2A) or sensitive (MCF-7:5C) to E2-induced apoptosis. To identify genes associated with E2-induced apoptosis, estrogen deprivation-resistant/apoptotic-sensitive 5C cells were compared to both estrogen-dependent MCF-7:WS8 and estrogen deprivation/apoptotic-refractory MCF-7:2A cells
