Transcriptional profiling of Schistosoma mansoni following knocking down Krüppel-like factor 4 (klf4) [RNA-Seq]

(Submitter supplied) klf4 is enriched in a putative tegument associated lineage of S. mansoni. To investigate the function of klf4, we compared transcriptional profiles of klf4 (RNAi) worms to those of control (RNAi) worms.

Transcriptomics of the racematic Praziquantel: R-PZQ versus S-PZQ in male Schistosoma mansoni

(Submitter supplied) Schistosomiasis affects over 250 million people worldwide and is caused by trematodes of the genus Schistosoma including the species Schistosoma mansoni. Praziquantel (PZQ) is the most widely available and implemented drug for this disease, and is produced as an racematic mixture composed of the enantiomers, R-PZQ and S-PZQ. In this study we examined the gene expression profiles of 49 day male S. mansoni after 18 hour treatment with R-PZQ, S-PZQ or control (1% DMSO). more...

Long non-coding RNAs are markers of Schistosoma mansoni single-cell populations

(Submitter supplied) Schistosoma mansoni is a flatworm that causes schistosomiasis, a neglected tropical disease that affects over 200 million people worldwide. New therapeutic targets are needed with only one drug available for treatment and no vaccine. Long non-coding RNAs (lncRNAs) are transcripts longer than 200 nucleotides with low or no protein-coding potential. In other organisms, they have been shown as involved with reproduction, stem cell maintenance and drug resistance, and they tend to exhibit tissue-specific expression patterns. more...

Rhesus macaques self-curing from a schistosome infection can display complete immunity to challenge

(Submitter supplied) The rhesus macaque provides a unique model of acquired immunity against schistosomes, which afflict >200 million people worldwide. By monitoring bloodstream levels of parasite-gut derived antigen, we show that from week 10 onwards an established infection with Schistosoma mansoni was cleared in an exponential manner, eliciting resistance to reinfection. Secondary challenge at week 42 demonstrated that protection was strong in all animals and complete in some. more...

A male-derived non-ribosomal peptide pheromone controls female schistosome development II

(Submitter supplied) βAlanyl-tryptamine (BATT) is a peptide secreted by male S. mansoni upon pairing with a female parasite. Treatment of synthetic BATT on virgin females wihtout any pairing is sufficient to induce sexual development and egg depostiion.

The Schistosoma mansoni nuclear receptor FTZ-F1 maintains esophageal gland function via direct transcriptional regulation of meg-8.3

(Submitter supplied) The schistosome homolog of ftz-f1 is a nuclear receptor. In order to understand the function of Smftz-f1, we performed RNAseq with samples taken on two days, Day 5 and Day 9, on either control (RNAi) or Smftz-f1 (RNAi) parasites to see the transcriptional consequences of Smftz-f1 RNAi and to find potential transcriptional targets.

A male-derived non-ribosomal peptide pheromone controls female schistosome development

(Submitter supplied) gli1 is a gene in the male S. mansoni required for female sexual development. As a putative transcription factor, RNAseq comparing pairs with gli1 knocked-down or control males was performed to look for the genes which are trancriptionally regulated by gli1. sm-nrps was one of the essential candidate discovered in this analysis that has been characterized in the same publication, revealing a pheromone directed novel mechanism of female schistosome sexual development.

Characterization of schistosomal microRNA by species, sex and developmental stage

(Submitter supplied) To date, 115 miRNAs genes (precursors) have been registered in miRBase for Schistosoma mansoni, and 56 for Schistosoma japonicum (225 and 79 mature miRNA, respectively). miRNAs have not been systematically described in Schistosoma haematobium. Our aim in this study was to systematically characterize and quantify miRNA in Schistosoma by species, developmental stage and sex and to test the usefulness of this characterization in biomarker discovery for infection. more...

Single-cell analysis reveals regulation of germline stem cell fate in the human parasite Schistosoma mansoni

(Submitter supplied) Schistosomes are parasitic flatworms that cause one of the most prevalent infectious diseases from which millions of people are currently suffering. Here, we construct a cell type atlas of juvenile schistosomes to capture germline stem cells (GSCs) in the course of de novo gonadal development.

Large-scale RNAi screening uncovers new therapeutic targets in the human parasite Schistosoma mansoni

(Submitter supplied) stk25 and tao genes came out of an RNAi screen in the parasitic flatworm Schistosoma mansoni, and these genes shared a similar knockdown phenotype. Transcriptomic analysis of reduced sensitivity to praziquantel in Schistosoma mansoni

(Submitter supplied) Schistosomiasis is a chronic neglected tropical disease caused by digenetic parasitic flatworms of the genus Schistosoma. The disease is estimated to affect over 206 million people, the majority of whom live in Africa where Schistosoma mansoni and Schistosoma japonicum are the major causative agents. While a number of drugs have been used to treat schistosomiasis, praziquantel (PZQ) is the only one that is widely available, relatively cheap, and easy to use. more...

Single-cell RNA sequencing of proliferative stem cell population from juvenile Schistosoma mansoni worms

(Submitter supplied) The rise of single-cell RNA sequencing (scRNAseq) technologies has enabled researchers to classify cell types, delineate cell developmental trajectories, and measure molecular responses to external perturbations. These technologies all rely on the premise that cell-to-cell variations arising from the biological processes of interest are clearly distinguishable from the intrinsic transcriptional and technical noise. more...
