Summary |
Escherichia coli release Extracellular Vesicles (EVs) which carry diverse molecular cargo. Pathogenic E.coli EVs contain virulence factors which assist during infection in the host in different mechanisms.The RNA cargo of E.coli EVs has not been assessed in their effect in the host. We used microarray data to asses and compare the global response of bladder cells to EV-RNA from pathogenic E.coli (Uropathogenic UPEC 536) and non-pathogenic E. coli (probiotic Nissle 1917)

Overall design |
EVs were isolated from RPMI-grown bacterial cultures, following centrifugation of spent medium, filtration, concentration and ultracentrifugation. Pelleted EVs were further purified using Size Exclusion Chromatography columns. RNA was extracted from pure EV preparations using a commercial kit and RNA quantified by Qubit. In vitro cultured bladder cells (5637) were treated with EV-RNA for 4 hours, after which cells were washed, removed and kept in TriZol at -80C until processing. RNA was extracted from bladder cells using a commercial kit and RNA quantified by Qubit, before determining the gene expression response by microarray technology.
